Adding a polypeptide, small protein, or enzyme to a gene of interest, also referred to as protein tagging, is a powerful and versatile technique widely used in biochemistry and molecular biology. In practice, an epitope tag is cloned in-frame relative to the gene of interest, either at the amino (N) or carboxy (C) terminus of the gene. As a result, a fusion protein is synthesized with the peptide tag.
